Nancy Guthrie, 84, disappeared from her home in Tucson on the night of 31 January or in the early hours of 1 February.
Authorities believe she was taken against her will, after blood found on her front porch was confirmed to be hers.
In a video posted to Instagram today, Savannah Guthrie said her family had reached what she described as “an hour of desperation”.
“I just want to share a few thoughts as we enter another week of this nightmare,” Guthrie said in the video, appearing alone, in contrast to previous recordings where she has appeared with her siblings.
“She was taken, and we don’t know where. And we need your help,” Guthrie added.
She thanked the public for their prayers, saying that her mother is also feeling them “even in this darkest place”.
Guthrie also urged anyone who had seen or heard anything unusual, “no matter where you are”, to report it to law enforcement.
The latest appeal comes as investigators continue to examine a series of alleged ransom messages sent to local and national media outlets.
At least one note demanded payment in Bitcoin and set out deadlines, some of which have now passed.
The FBI has said it is reviewing the messages, but has not confirmed their credibility.
The family previously indicated they were willing to pay for Nancy Guthrie’s safe return and released several videos addressing her potential captors directly, asking for proof that she is alive.
Law enforcement officials say no suspects or vehicles of interest have been identified.

Surveillance cameras at the property failed to capture usable footage after a doorbell camera was disconnected shortly before Nancy Guthrie vanished.
The case has drawn widespread attention across the US, with extensive media coverage and heightened law enforcement presence in the quiet Tucson neighbourhood where she lived.
The FBI is offering a $50,000 (€42,000) reward for information leading to her recovery.
Officials have expressed increasing concern for the missing woman’s health.
She is understood to have heart problems, uses a pacemaker and requires daily medication.
US President Donald Trump has said federal authorities are assisting in the investigation, describing it as “going very well” and suggesting strong leads may emerge.
